Then, we designed three cDNA chains matching the target in varying degrees and probed into their advantages, respectively. As a result, a new cDNA chain, called cDNA-4, was produced with a part of base sequences optimized. Through the performance analysis of the G-quadruplex, I proposed to replace the STP-Probe chain with a new-found chain named 22-AG because of its more vigorous fluorescence intensity.<strong><span style="color: #1dadff;">【分层次陈述研究中的几个关键步骤】</span></strong>The biggest challenge of our research was that the testing data of fluorescence intensity was unstable. Hence, changing the experimental conditions, we set several control groups for comparison, summarized the similarities of experimental data acquired in different turns, and got conclusions. The bonding strength between the target chain and the cDNA chain was stronger than that between the Probe-chain and cDNA chain. The annealing of the DNA solution was crucial to reinforce the triple helix between the cDNA-4 chain and the STP-Probe chain. In the subsequent experiment, we found the root cause for the unstable data&mdash;the light source for a fluorescence spectrophotometer had been used for too long, resulting in the instability of the fluorescence intensity. We took the G-quadruplex/ heme enzyme complex as the second output signal and carried out the colorimetric detection. Hard work pays off. We illustrated the bonding strength between the 22-AG chain and ThT by the fluorometric/colorimetric dual-signal test, thereby proving the strong bonding strength between the target chain and cDNA chain.<strong><span style="color: #1dadff;">【描述遇到的困难及其解放方案和最终结果】</span></strong>Lasting for one year, this research project was indeed a huge challenge for us. Despite that, this research deepened my understanding of abstruse chemical principles and enabled me to apply knowledge more flexibly, ponder issues more thoroughly, and conduct experiments more rigorously.
